Title Post-transcriptional regulation by small RNAs in Hevea

Description Role of small RNA on the regulation of genome expression has been intensively studied in model plants. By contrast, little is known on perennial plants but a few studies revealed specific small RNA-mediated posttranscriptional regulations. We completed the recent release of massive genomic, transcriptomic and micro-transcriptomic data by a degradome analysis in order to identify all mRNAs cleaved by small RNAs. This work revealed that latex cells had a high small RNA-mediated transcript cleavage activity by miRNA and phased siRNA compared to other tissues. Post-transcriptional regulation was observed at each step of the natural rubber biosynthesis pathway. The absence of a conserved pathway for ta-siRNA production as well as partial conservation of targets revealed a divergence in interspecific post-transcriptional regulation suggesting a possible role during plant speciation and adaptation to environmental stresses. This comprehensive analysis of posttranscriptional regulations at the genome-wide level revealed new insight into evolutionary mechanisms of RNA silencing
