Description In this study, a nested design experiment was conducted in a semiarid temperate steppe in northern China and totally 705 topsoil samples were collected in five sampling quadrats. We focus on a small scale (1.19 m2 for a quadrat) with easy to thoroughly sampling, to illustrate how many prokaryotic species inhabit in 1.19 m2 grassland topsoil (0-20 cm), and to assess the effects of sampling strategies, including sample numbers, biological or technical replicates and their combinations, pooling and sequencing depth on the captured and estimated richness, as well as widespread common species.
