Description **** The processed data is accessible through OMIX894 and OMIX895.**** Fetal liver is the site of hepatogenesis and hematopoiesis and contains many cell lineages derived from the endoderm and mesoderm. However, the characteristics, developmental programs, similarities and difference from human to mouse of many cell lineages in fetal liver remain unclear. We performed single-cell RNA sequencing of human 5-19wpc (W5-19) and mouse E11.0-17.5 whole fetal livers. We identified endoderm-derived, mesoderm-derived, and hematopoiesis-derived 13 major cell types and defined the developmental pathways of these cell types. We identified novel markers for hepatic lineages and an ID3+ subpopulation of hepatoblasts in human and mouse, and verified that the differentiation of hepatoblasts obeys the default-regulatory model. We found that human but not mouse fetal hepatocytes displayed heterogeneity. During human and mouse hematopoiesis, we detected the maturation process of erythroid progenitor cells. Moreover, we observed different cell lineage compositions during hematopoiesis in the human and mouse fetal liver. In all, we revealed the dynamic cell landscapes of fetal liver and determined the similarities and differences in liver development between species, which provides extensive resources for inducing various liver cell lineages in vitro.
