Description Using CRISPR-Cas9 to disrupt the CHD8 gene in the embryos of cynomolgus monkeys, we found that CHD8 mutations selectively increased gliogenesis rather than neurogenesis to cause macrocephaly and autism-like behaviors in monkeys. Our findings support the idea that gliogenesis is critical for brain size in the primates and suggest that abnormal gliogenesis may result in ASD.
