Cystic Echinococcosis (CE), caused by infection with larval stage of tapeworm Echinococcus granulus sensu lato (E. granulosus s.l.), represents a near-cosmopolitan zoonotic disease. Various infiltrating immune cells gather around the lesion and produce a lesion microenvironment, but the unique composition and cellular heterogeneity in hepatic CE lesion microenvironment are incompletely understood. Here, 88020 immune cells isolated from peripheral blood (PB), adjacent normal (AN) liver tissue and peri-lesion liver (PL) tissue from four CE patients were profiled using single-cell RNA sequencing (scRNA-seq). We identified 23 discrete cell populations, and there existed distinct differences in infiltrating immune cells between tissue environment. Despite significant similarity between PL and AN tissue-resident immune cells, cellular proportions of innate lymphocyte 2 (ILC2) and plasmacytoid dendritic cells (pDC) were higher in PL tissue. Interestingly, NFKBIA gene was up-regulated in these phenotypes. For CD4+ T cell populations, nine cell subsets were found, and Th2-CD4+ T cells were significantly increased in PL tissue. There existed close contact among CD4+ T cells with ILC2 and pDC cells, which caused upregulation of genes related to positive immune activity in AN tissue. However, genes related to immunosuppression especially the immune inhibitory checkpoint NKG2A/HLA-E were obviously increased in PL tissue, suggesting an inhibitory state of CE lesion microenvironment resulted from cellular interaction. This work offers new insights into transcriptional heterogeneity of infiltrating immune cells in hepatic CE lesion microenvironment, providing significant implications for potential target signatures for diagnosis and immunotherapies. |
