Title Flower Bud Differentiation Transcriptomic Analysis of the Lateral-flowered Tree Peony Cultivars

Description Objective:Analyze the transcriptomics of different flower bud differentiation stages of tree peony varieties,and explore the internal molecular mechanism of hormone metabolism related regulation of flower bud differentiation,so as to provide reference for the research on flower bud differentiation mechanism and the molecular mechanism of floral formation of tree peony cultivars.Method: Illumina high-throughput sequencing technology was used for transcriptome sequencing of flower buds of three stages: primordium undifferentiated stage (LUP),lateral flower primordium differentiation initial (LIP) and lateral bud differentiation stage (LDP),and bioinformatics methods such as gene function annotation and differential expression gene screening were used to analyze the high quality sequences obtained by sequencing.Results:A total of 23.48 G high-quality data were obtained by transcriptome sequencing,and 207,590 Unigenes were assembled by Trinity software.A total of 103 155 unigenes were finally obtained with functional annotations against the 7 databases.We compared gene expression levels among samples at LUP,LUP and LIP,with LUP as the control. Furthermore,37,407differentially expressed genes (DEGs) were obtained,in details,19,256 DEGs were up-regulated while 21,230 DEGs were with down-regulated expression. Based on GO enrichment and KEGG pathway enrichment analysis,33 genes related to the formation and development of lateral buds in tree peony were screened.Of these,19 were closely related to hormonal response,and 14 were related to branching,regulation of lateral budding, and lipid synthesis of unicorns.
