Description we generated a de novo cynomolgus macaque genome assembly(MaFaC) using long-reads sequencing technology(Pacbio,Bionano,HI-C) and multiple scaffolding strategies. Compared to most frequently used NGS cynomolgus macaque reference genome(MF 5.0), in our assembly, N50 reaches 6.03Mbp which improved 75 folds in terms of length, 76119 gaps(137.6 Mbp) were closed and Y chromosome was included. Also, we provided gRNA and wild mutants spectrum of cynomolgus macaque to facilitate its usage as an non-human primate model in Cas9-gRNA gene editing . Furthermore, taking advantage of MaFaC long reads data, we identified much more novel structure variations to be a genetic resources for future studies of primate genetics, evolution and diseases.
