This project first conducts single-cell transcriptome sequencing (scRNA-seq) of human oligodendrocyte precursor cells to study the heterogeneity of their cell pairs. Preliminary results found that human oligodendrocyte precursor cells contain two cell subpopulations, one is a cell population dominated by PDGFR-α and CNPase, accounting for 90.56%, while the remaining part has low expression of OPC-related indicators, belonging to Non-OPC group. We will continue to study the differences in the nature and functions of these two groups of cells from the aspects of gene expression differences, GO function and KEGG pathway analysis. In addition, we perform transcriptome sequencing (RNA-seq) on neural stems and different generations of OPC. The results showed that with the increase of OPC generation, the markers related to neural stems, stellate keratinocytes, and neurons showed a downward trend. The expression of OPC markers is higher than that of NSC, such as PDGFR, OLIG1/2, NKX2.2 and A2B5. |
