描述信息 Given our increased awareness of genomic diversity, more high-quality reference genomes are necessary for understanding human evolution and genome complexity. Here, we produced 35 high-quality genomes of diverse humans from across the world assembled by more than 50x long reads complemented with short-read sequencing. 283.8 Mb of novel sequences were not present in the reference genome, particularly in Africans, who displayed larger genome size and more novel sequences. The assemblies facilitate investigations on evolutionary dynamics of the largest gene family, i.e., olfactory receptors across diverse human populations. Africans and non-Africans exhibited differentiation of olfactory receptor patterns, while Asians displayed higher olfactory sensitivity than Africans. Comparative population genomics identified structural variants with population differentiation between African and non-African populations, possibly causing different phenotypes and incidences of certain diseases. Finally, by mapping to these de novo genomes, we categorized insertions introgressed from archaic hominins. This study presents comprehensive genome data and diverse assemblies, which should help improve our understanding of human evolution and could be applied in future genetic studies.
