Description Low-input Hi-C can be performed with as little as a single mature leaf of Arabidopsis (~0.1g fresh or frozen tissue), compared to the 10 grams of fresh tissues (or 2 g fixed tissues) required for the regular Hi-C in Arabidopsis (Feng et al., 2014; Grob et al., 2014; Wang et al., 2015). The key changes in our low-input Hi-C protocol involves crosslinking of isolated nuclei (instead of crosslinking fresh tissue), in-nuclei ligation, and using Tn5 tagmentation to combine fragmentation and library construction into one step.
