描述信息 This study examines the molecular characteristics linked to the efficacy of immunotherapy in gastrointestinal (GI) cancers by analyzing whole-exome sequencing data from the JUPITER-06 cohort and RNA sequencing data from the Capability-01 cohort. JUPITER-06 Cohort: Patient data were sourced from the JUPITER-06 prospective clinical trial, which evaluated the efficacy of chemotherapy in combination with the PD-1 antibody Toripalimab. Formalin-fixed paraffin-embedded (FFPE) tumor tissues and peripheral blood samples (for germline control) were collected prior to the initiation of therapy. Genomic analyses were performed on the collected tissue samples. This cohort encompasses 95% of the total patient population enrolled in the JUPITER-06 trial. Capability-01 Cohort: Patient data were obtained from the Capability-01 prospective clinical trial, which assessed the efficacy of a novel triplet therapy comprising a PD-1 antibody, the HDAC inhibitor Chidamide, and the VEGF antibody Bevacizumab. To elucidate the underlying mechanisms contributing to the clinical effectiveness of this triplet therapy, paired biopsy samples of tumor tissue were collected both prior to treatment (n = 10) and post-treatment (n = 10). RNA sequencing was performed on these samples, with data derived from the doublet arm including 6 patients and from the triplet arm including 4 patients.
