描述信息 Here we report a novel laboratory mouse model derived by engineering that has fewer chromosome pairs than its wild-type counterpart. The engineering was initially performed within embryonic stem cells carrying a simplified haploid karyotype, in which nonhomologous chromosome arms could be ligated simply after near-ending DNA cleavages. By differentiating haploid stem cells into diploid somatic cells, we found an upper-length-limit for ligated chromosome arms, exceeding which would result in cell polyploidization. By oocyte injection of haploid stem cells, full-term pups carrying ligated chromosomes with arm sizes beneath the upper-length-limit were derived, which could transmit ligated chromosomes to their offspring. Our work achieves chromosome-level rearrangements common in evolution and disease in house mouse, and exhibits a feasible route for performing large-scale DNA engineering in mammals.
