To investigate how PAK2 affected neural tube closure by regulating gene expression, RNA-seq was performed on WT (3 embryos in one replicate, 3 replicates) and Pak2 knock out (3 embryos in one replicate, 2 replicates) bulk embryos at E9.5. To clarify cell-specific abnormality in the Pak2 knock out embryo, we performed scRNA-seq by using the DNBelab C4 platform. A total of ~48 000 cells that passed quality control in WT and Pak2 knock out embryo were divided into seventeen clusters with t-distributed stochastic neighbor embedding (tSNE). |
