To generally investigate how the inhibition or knockout of SQLE affect β-glucan induced trained immunity, we had processed RNA seq after first stimulation and secondary stimulation, and ATAC-seq before secondary stimulation. RNA-seq after first stimulation could reflect the change of genes and pathways after β-glucan training or SQLE inhibition or deficiency. These changes would trigger the subsequent metabolic changes and epigenetic rewiring. ATAC-seq before secondary stimulation revealed the chromatin accessibility of genes related to inflammatory response, immune response and metabolic process. The accessibility would determine the intensity of secondary response. Thus, RNA-seq after secondary stimulation would verify the status of trained immunity. |
