Description We found NOVA1, a neuron-specific RNA-binding protein, was specifically upregulated during the adipogenic induction of human white adipose tissue derived stromal/stem cells (hASCs). To explore its molecular roles in adipogenesis, hASCs were infected with shNOVA1 or the control lentiviral particles and then adipogenically induced for 3 days. The gene expressions of the cells were profiled using high density Affymetrix Human Transcriptome Arrays (HTA 2.0) and RNA-Seq. CLIP-Seq was applied to detecting the NOVA1-RNA interaction during white adipogenesis. ATAC-Seq was also performed to observe the effect of the interaction between NOVA1 and its target NCOR2 on the chromatin accessibility of hASCs.
