描述信息 Wild bananas are famous for being processed as industrial products like natural fibre, as well as their nutritive value. However, their genetic diversity has not been fully explored. In this study, we sequenced and assembled two representative Australimusa bananas genomes, Musa textilis Abaca with high quality natural fiber and Musa troglodytarum Utafun with abundant β-carotene, which both possess ten pseudochromosomes with a total size of 613 Mb and 619 Mb, respectively. Comparative genome analysis of the two genomes, together with other reported genomes from Musaceae family, provided insight into Musaceae evolution history and genome size variation driven by unequal recombination mediated transposons removal of Musaceae. Compared with other wild bananas and Agava sisalana genomes, the mature pseudostem of Abaca enrich cellulose, lignin, hemicellulose and depress pectin content contributing to thicker cell wall and superior mechanical properties. The expression of cellulose-synthesis genes peaked earlier in Abaca than in A- and B-genome during growth development, possibly resulting in an earlier accumulation of cellulose at secondary cell wall (SCW) formation. We screened some candidate cellulose- and lignin-related genes involved in fiber biosynthesis and validated some transcription factors regulating key candidate genes to elucidate the genetic mechanisms underlying the superior fiber properties of Abaca. The Abaca-specific expressed gene MusaMYB26, which is directly regulated by MusaMYB61, may be an important regulator promoting precocious expression of the SCW MusaCesAs. Overall, the high-quality reference genomes assembled in this research may contribute to understand the genetic diversity of Musaceae family and provide genetic basis for functional genomic studies in bananas.
