Description |
DNA organization, replication and transcription and the related regulations are key life processes for genome integrity and the transmission of genetic information. Multiple protein machineries consume ATPs to execute these life processes, whose dynamics and kinetics will reflect their biological functions. In this subproject, we aim to investigate the interactions between protein machineries and genes at single-molecule level. To achieve this, we focus on the development and application of novel single-molecule techniques to study the dynamic organization and regulation of chromatin DNA, underscoring the mechanism for the interactions between SWR1 complex and H2A nucleosomes, especially uncovering the mechanism of the regulation of the ubiquitination on the dynamic structures of nucleosomes; discovering the mechanisms of the interactions between helicases and DNA substrates involved in the processes of DNA replication and repair; underscoring the mechanisms of Rob and SoxS factors on transcription initiation. These results are highly relative with other subprojects in project one, which promote the technical combinations of structure analysis with X-ray/Cryo-EM, single-molecule analysis with sub-nanometer resolution and molecular dynamics simulation to underscore the dynamic features and mechanisms of biomolecular complexes and processes deepening our understandings on the mechanisms of genome maintenance and transmission. |
