描述信息 Cell fitness is one of the most vital features that endows the cell graft with the ability to adapt to its new environment, contribute to tissue function and maintain bodily homeostasis. In this proof-of-concept study, we achieved a safe and cost-effective manipulation of cell fitness in a typical less-fit cell type (human chondrocyte) for tissue regeneration, using a chemical compound FPH2. FPH2 was identified to significantly improve and maintain chondrocyte phenotypes under different challenging conditions. Using single-cell transcriptomics, we discovered that FPH2-driven winner status was a result of chemically programmed gene expression associated with energy metabolism, protein synthesis, cell proliferation, cell motility and self-protection. This study proposes an applicable chemical approach to drive cell fitness for tissue regeneration, and provides mechanistic insight into the underpinning molecular machinery.
