This project will study the structure of H2A variant nucleosome which is significantly different from the conventional nucleosome structure, as well as the complex structure formed by chromatin modifying enzymes, chromatin remodeling complex subunits and nucleosome, comprehensively using structural biology and biochemistry methods such as cryo-electron microscopy, X-ray crystallography, monomolecular magnetic forceps. Our study aims to reveal the structural basis and molecular mechanism of variant nucleosome and nucleosome modification regulating chromatin dynamics, clarify their important roles in DNA replication, transcription, repair and other life activities, and explore potential drug targets with promising application prospects. |
