The RNA profiles of erythrocytes possess pathological features that could be used for early cancer detection. This study evaluated the diagnostic capacity of erythrocyte RNA profiles in a cohort of patients different types of cancers and healthy donors. We performed transcriptome sequencing for erythrocytes isolated from above 4,000 blood samples. Erythrocytes were isolated from 5 mL purple-capped vacutainers equipped with the anti-coagulant EDTAK2. The total RNA was purified with RNAprep Pure Micro Kit and then subjected to library preparation using NEBNext Ultra RNA Library Kit for Illumina or SMART-Seq v4 Ultra Low Input RNA Kit for Sequencing. Finally, the libraries were submitted for 150 bp paired-read sequencing on the Illumina Hiseq X-ten. |
