As the most abundant tissue in the body, skeletal muscle plays a crucial role in performing essential bodily functions. Pigs, being excellent biomedical models, are particularly valuable for studying skeletal muscle development due to the ease of obtaining sufficient skeletal muscle samples at various prenatal and postnatal developmental stages, as well as individuals with essentially similar genetic backgrounds. However, the dynamic landscape of super-enhancers (SEs) and their regulatory networks during skeletal muscle development remains unclear. This study aims to elucidate how SE-mediated transcriptional regulatory mechanisms drive pig skeletal muscle (PSM) development. To achieve this, we employed the Cleavage Under Targets and Tagmentation (CUT&Tag) technique to profile the H3K27ac landscapes and identify SEs in PSM samples from five developmental stages. These stages encompassed prenatal myofiber formation to postnatal myofiber hypertrophy. Through our analysis, we discovered a subset of SEs that significantly contribute to muscle development and traits associated with pig meat and carcass. Using an innovative approach that integrates information from eRNA annotation, GWAS signals, and high-throughput capture Self-transcribing Active Regulatory Region Sequencing (STARR-seq) experiments, we present potential critical enhancers and their regulatory networks that might potentially play important roles in skeletal muscle development and growth in mammals. |
