描述信息 Breast cancers present intricate microenvironments comprising heterotypic interactions between different cells. A global map outlining the cellular organization of breast cancer has yet to be established. Here, we leverage the DNA nanoball-based genome-wide in situ sequencing (Stereo-seq) to visualize geospatial architecture of thirty primary breast tumors and metastatic lymph nodes. The unprecedented single-cell resolution mapping enables accurate localization of various cell types within the tumor ecosystem and discerns the delicate structure of sporadic vasculature. In particular, we unveil the immunomodulatory function of venules, in which ACKR1+ venular endothelial cells and CCL21+ venular smooth muscle cells constitute a pro-inflammatory niche to synergistically promote immune cell chemotaxis and extravasation into tumors. High numbers of venules are associated with more immune infiltration and better clinical outcome of patients. This study represents currently the most comprehensive survey of the spatially resolved atlas of human breast cancer across four molecular subtypes and provides critical insights into the phenotypic and spatial heterogeneity of tumor vasculature.
