IFNγ-induced immunoinhibitory feedback is a crucial mechanism for endothelial cells to perform immunoregulation of T cells. However, whether TNFα is involved in regulating this program is unclear. Mouse vascular endothelial cells sEND.1 was taken for invitro experiment. After several passages and exclusion of mycoplasma contamination, sEND.1 cells were divided into two groups, including three samples in each group. One was treated by IFNγ (20ng/ml) as the control group (IFNγ), and another one was administrated with a combination treatment of TNFα (10ng/ml) and IFNγ (20ng/ml) as the experimental group (TNFα-IFNγ). 24h later, RNA was extracted from samples and sent to BGI for subsequent deep sequencing. |
