While the clinical importance of the periosteum in bone growth and repair has been widely acknowledged, the precise identification and understanding of the role of periosteal stem cells (PSCs) in bone repair and regeneration have proven elusive. In this study, we employ single cell sequencing technology combined with a swine model to identify stem cells in the calvarial periosteum for the first time. Our investigation identifies ALPL+PDGFD+ (AP+) cells as distinctive calvarial PSCs possessing both self-renewal ability and differentiation potentials. |
