Description To elucidate the somatic lineages and evolutionary dynamics in preneoplastic lesions, we established a transgenic mouse line carrying the lineage tracing barcode and used it to generate lineage traced neoplastic lesions using two mouse models (an azoxymethane (AOM) and dextran sodium sulfate (DSS) mouse model and an ApcMin/+ mouse model) of colorectal tumorigenesis. Through integrated analyses of single cell phylogenies (n=105 samples), whole-genome sequencing (n=31 mouse samples) and single-cell RNA sequencing (n=9 mouse samples), we have investigated the trajectory of intestinal tumorigenesis observed in both mouse models.
