Tumor endothelial cells (TEC) are the key component of the tumor microenvironment (TME), and endothelial-to-hematopoietic transition (EHT) is a conserved process for generating hematopoietic systems in the embryonic stage. However, it is unclear that whether hematopoiesis occurs within TMEs. Armed with mouse tumor and linage tracing dual-function models, a patient-derived tumor-like cell clusters (PTCs) model, and human clinical samples, here we report a new type of intratumor hematopoietic stem cell (iHSC) originated from in situ tumor-induced EHT in TMEs. We utilized various single-cell transcriptomic sequencing technologies, including STRT-seq, SmartSeq2, and 10x Genomics, to sequence samples from patients with colorectal cancer (CRC) and lineage tracing mouse models. |
