Title SETD1A/B regulates transcription-dependent H3K4me3 establishment and lineage segregation in mouse preimplantation embryos

Description To elucidate the specific histone methyltransferase accountable for H3K4me3 deposition in the early stages of mouse embryogenesis, we conducted H3K4me3 ChIP-seq analysis on Late2C and morula stage embryos following targeted knockdown of Mll2 and Setd1a/b. To examine the transcriptional impact on H3K4me3 dynamics, we performed Pol II and H3K4me3 ChIP-seq following a brief exposure to triptolide at intervals of 0, 2, 4, and 6 hours on embryos at the Late2C and morula stages, respectively. Furthermore, to explore the functional role of H3K4me3 during early embryonic development, we executed H3K4me3 ChIP-seq and RNA-seq analyses subsequent to either the knockdown or overexpression of enzymes responsible for the addition and removal of H3K4me3 marks, respectively.
