Title The waterlogging-induced gene ZmEREB179 acts as a negative regulator of waterlogging tolerance in maize seedlings

Description Maize (Zea mays) is highly susceptible to waterlogging stress, which reduces both the yield and quality of this important crop; thus, elucidating the molecular mechanisms governing waterlogging tolerance is a key research goal. In this study, we identified ZmEREB179, encoding an ethylene response factor (ERF) located in the nucleus, as being induced by waterlogging stress and ethylene. Overexpressing ZmEREB179 heightened maize sensitivity to waterlogging stress. Conversely, the Zmereb179 knockout mutants were more tolerant of this stress, indicating that ZmEREB179 functions as a negative regulator of waterlogging tolerance. A transcriptomic analysis of the ZmEREB179-overexpressing lines revealed that ZmEREB179 modulated the expression of various stress-related genes, including the previously cloned ZmEREB180. Transient and simplified cleavage under targets and tagmentation, electrophoretic mobility shift, dual-luciferase, and yeast one-hybrid assays verified that ZmEREB179 directly targets the ZmEREB180 promoter and represses its expression. Furthermore, association analyses between 220 maize inbred lines and their waterlogged phenotypes led to the identification of a ZmEREB179 promoter variant that was significantly associated with greater waterlogging resistance. This variant was verified in two F2 segregating populations, and holds promise for practical application in breeding programs. Our findings shed light on the involvement of the transcriptional cascade of ERFs in regulating plant waterlogging tolerance.
