Pre-harvest germination (PHS) is a serious threat to the yield and quality of global crop seed production due to the lack of seed dormancy. PHS and seed dormancy as complex quantitative traits. Current genetic studies or breeding of crops with suitable seed dormancy are limited by fewer effectively regulated genes. Through the isolation and cloning of maize vp16 mutation sites and candidate genes, we confirmed that the candidate gene encodes Zmccr3, which is involved in lignin metabolism and regulates seed germination and dormancy. However, the molecular mechanism of its regulation is still unknown. Therefore, this project aimed to confirm the weighted co-expression network (WGCNA) regulated by Zmccr3 and clarify the metabolic network of Zmccr3 regulating grain germination and dormanness by using transcriptome analysis at two developmental stages of grain with differential vp16 locus (BC4F2). Transcriptomics, proteomics, physiological and biochemical tests were used to verify the metabolic pathways regulating grain germination and dormancy at different stages of grain development with differential vp16 loci. The completion of the project will elucidate the molecular mechanism of ZmcCR3-mediated lignin metabolism in regulating seed germination and dormancy. It can provide new regulatory pathways and new genes for maize seed germination and dormancy, and provide gene resources for molecular breeding of PHS, seed germination and dormancy. |
