项目标题 Cryo-EM structure of the human subcortical maternal complex and the associated discovery of infertility-associated variants

描述信息 Maternal effect factors play an important role in the regulation of human egg maturation and early embryo development. In recent years, clinical studies have reported many maternal effect gene mutations closely related to female infertility. This project is aimed at patients with egg and embryo development disorders. Compared with patients with normal development of eggs and embryos. The maternal effector factor protein expression, DNA methylation, and metabolism levels of discarded eggs, embryos, follicle fluid, seminal plasma and embryo culture medium are detected. DNA/RNA sequencing, proteomics, and metabonomics technology are used to detect the changes in related factors. By combining the prediction of complex stability of maternal effector factors and the in vitro reconstitution, to provide methods for identifying pathogenic variants. Finally, to reveal the mechanism of maternal effect factors related infertility, and to explore the possible clinical treatment strategies.
