Epilepsy is a chronic disease caused by sudden abnormal discharge of brain neurons leading to temporary brain dysfunction, which seriously affects human physical and mental health of the nervous system disease, because of its repeated attacks and long duration, over the years to the patient's family and even the whole society has brought great pressure. At present, the treatment of epilepsy is mainly based on drugs, but because of its unknown pathogenesis, resulting in poor drug control effect. Since the ketogenic diet was first applied to epilepsy in 1921, it has been shown to be an effective therapy, but the molecular mechanism of the ketogenic diet's involvement in the treatment of refractory epilepsy remains unclear. From the perspective of nutritional metabolism, our study revealed that ketogenic diet regulates glutamate metabolism in mitochondria by increasing the expression of SIRT4, thus leading to the accumulation of inhibitory neurotransmitter GABA, which plays a role in slowing seizures. This study is of great significance for the clinical scientific use of ketogenic diet and the improvement of the cure rate of epilepsy. |
