Shengxian Decoction (SXD) is a Traditional Chinese Medicine formula for improving the cardiovascular diseases and fatigue caused by chemotherapy in clinic. However, the detailed therapeutic evaluation and mechanism of SXD in regulating immunosuppression are still unclear. Here we demonstrated that SXD treatments predominantly improved B cell immunity in immunocompromised mice induced by cyclophosphamide (CTX). SXD partially reversed the CTX-induced reduction of mouse spleen T cells and B cells. RNA sequencing revealed that SXD mainly promoted B cell-mediated immune response and B cell receptor pathway. Among 202 up-regulated genes, SXD increased the expression of transcription factors Pax5, Tcf3 and Myc, which were essential for B cell proliferation and differentiation. In addition, SXD significantly increased the level of class switched antibodies and IgA+ B cell proportion. Taken together, this study demonstrates a protective effect of SXD against CTX-induced immunosuppression by up-regulating B cell immunity. |
