Description A proximity-dependent biotin identification (BioID) assay was used to identify the interactome for Musclin on the plasma membrane of adipocytes. pCMV-BirA* (Control), pCMV-Musclin-BirA* plasmids were transfected into the HEK293T cells. After transfection for 12 h, cell medium-enriched in BirA* or Musclin-BirA* fusion protein were collected for incubation of C3H10T1/2-derived adipocytes, supplemented with 80 μM biotin for 24 h. The membrane-associated proteins from adipocytes were separated and were then pulled down using streptavidin agarose. The agarose beads were pelleted for obtaining Musclin-interacted proteins, followed by denaturation, silver staining and M.S. analysis.
