Description |
Proteins immunoprecipitated from SDS-PAGE gels were digested as previously described. The gel pieces were washed with 50% ACN for 30 min and then with 100% ACN for 30 min. Following reduction and alkylation, the proteins were digested with trypsin at 37 °C for 12 h. The supernatant containing the digested peptides was collected and processed for desalination using an HPLC buffer. The samples were analyzed by LC-MS/MS using an Easy-nLC1200 chromatograph (Thermo Scientific) and a Q Exactive plus mass spectrometer (Thermo Scientific). Furthermore, data analysis was performed using the predicted sequence of the NSD1-916aa protein and by searching against the UniProt-reviewed Homo sapiens database with Proteome Discoverer 2.4. |
