This study reports the highly efficient generation of blastoids from induced pluripotent stem cells (iPSCs) and somatic cell nuclear transfer ESCs (nt-ESCs) obtained from aged monkeys. With our protocol we achieve a blastoid formation efficiency of up to 80%. Additionally, we developed a hydrogel-based microfluidics platform to produce size-adjustable, biodegradable and monodisperse blastoid capsules in a scalable and reproducible manner. The capsules offer a stable 3D structure and mechanical protection during delivery through fallopian tubes. The high efficiency and scalable production of monkey blastoid capsules from reprogrammed aged somatic cells provides a powerful tool to advance the studies of embryonic development and may have broader implications for regenerative medicine. |
