Description | We have de novo design, synthesis, and shuffle a synthetic Igh locus (SynIgh). Compared to the natural Igh locus, the SynIgh (1) features a fully modular, bottom-up hierarchical design based on studies of natural loci; (2) incorporates V, D, and J genes from diverse global human populations; and (3) includes an inducible synthetic genomic rearrangement system. We then generated a SynIgh mouse model and test its function, including V(D)J recombination, support B cell development, and produce BCRs. We further investigated the capacity of synthetic rearrangement to shuffle the SynIgh locus in vivo, and assessing its impact on gene utilization and overall V(D)J recombination patterns. | ||||||||
