描述信息 Intratumoral heterogeneity (ITH) has been investigated primarily in locally advanced or metastatic cancer; however, much less is known about ITH in early-stage cancer, and the origins of ITH are poorly understood. We performed single-cell transcriptome profiling of early-stage lung adenocarcinoma from 12 patients, constructed a map of 88,695 cells, profiled the transcriptome states of tumor cell populations, and incisively explored the ITH of neoplastic cells. Significant transcriptional ITH is already present in early-stage lung adenocarcinoma (12 of 12 patients). The observed heterogeneity originates from two main sources. On the one hand, during tumor evolution, epithelial dedifferentiation can serve as a predictor of prognosis. On the other hand, early-stage tumor cells retain the original AT2 lineage differentiation program, representing an intermediate transition state from AT2 to AT1 cells. Two key transcription factors KLF4 and JDP2 were identified, overexpression of which in late-stage LUAD reverts tumor cells to this intermediate transitional state.
