| 描述信息 | To analyse the morphology, pathogenicity and drug resistance phenotypes of more than 2000 representative pathogenic fungal strains of Candida, Cryptococcus, Aspergillus fumigatus and Trichoderma viride. Cellular and colony morphology of the representative strains were analysed by using synthetic media and simulated host microenvironmental conditions, and a complementary animal infection model was developed to detect pathogenicity, and an in vivo and ex vivo model was developed to detect the drug resistance of the clinical strains. Through integrated analyses, the link between morphogenesis and pathogenicity and drug resistance was established. Using second-generation sequencing technology, genome sequencing will be carried out on more than 2,000 representative strains, and third-generation genome sequencing will be carried out on some strains with multi-drug resistance or high pathogenicity. We will analyse the relationship between genotypes, genome sequence variation and recombination and pathogenicity and drug resistance, as well as analyse the pattern of their evolution, so as to reveal the virulence and drug-resistance of the most important pathogenic fungi in China. Focusing on multiple signalling and metabolic pathways such as sterol synthesis and cell wall synthesis, we will focus on the key resistance regulators such as target enzyme coding genes of azoles and echinocandins, multi-drug resistance genes and PKC pathways, and analyse the resistance mechanisms of clinical strains of fungi. | ||||||||
