In this project, we demonstrated that FASN is essential for leukemogenesis. To investigate the role of FASN in leukemia progression, we established MOLM-13 cells with FASN knockdown. The cells were collected after selection with 1μg/mL puromycin for two days. Additionally, we treated MOLM-13 cells with a FASN inhibitor, MS-C19 (5μM), for 48 hours. The samples were collected for RNA sequencing, label-free quantitative proteomics, and lipidomics analysis. By performing multi-omics analysis on FASN-depleted and control MOLM-13 cells, as well as MS-C19 treated and untreated cells, we demonstrated that both MS-C19 treatment and FASN deficiency triggered the activation of lysosomal and inflammatory gene expression. |
