Alzheimer's disease (AD) features progressive amyloid deposition and cognitive decline. Forty hertz (Hz) light treatment has been shown to improve cognitive function, but its mechanisms and efficacy with concomitant AD therapies remain underexplored. This study examined the effects of 14-day low-intensity 40 Hz blue light exposure on cognitive functions and pathological changes of 5xFAD mice at different ages. The treatment prevented memory decline in 4-month-old 5xFAD mice and motivation loss in 14-month-old 5xFAD mice, together with restoration of glymphatic drainage and AQP4 polarity in memory-related regions. Importantly, the improvement required glymphatic drainage and activation of the vLGN/IGL-Re circuit. Furthermore, the combined use of 40 Hz blue light with anti-Aβ antibody significantly enhanced soluble Aβ clearance and cognition in 5xFAD mice. These findings provide new insights into the therapeutic mechanisms of 40 Hz blue light in mouse model of AD-related pathology, suggesting potential clinical benefits of light therapy alongside anti-Aβ antibody therapy. |
