Nuclei isolated from 50,000 counted cells were used for transposition reaction with Tn5 transposase (Vazyme Biotech). 10 PCR cycles were performed for library setup for each reaction mix using high-fidelity 2x PCR Master Mix (New England Biolabs) using primers with unique barcodes (Vazyme Biotech). Pair-end sequencing (150bp x 2) was performed on Illumina HiSeq X Ten instrument (CloudHealth Genomics Ltd.) |
