描述信息 Lentiviral vector transfection is an effective means to develop stable cell lines to express external genes. One question that needs to be explored is how external lentiviral vectors and overexpressed genes change the translation dynamics of cell lines. In this study, we constructed two transfected cell lines from human embryonic kidney 293T cells through lentiviral vector transfection. RNA sequencing was performed, followed by the identification of differentially expressed genes (DEGs) and genes with differential transcript usage (gDTUs) across cell lines. It was found that enhancing the expression of the green fluorescent protein reporter gene resulted in alterations to the expression of merely 31 genes, while hundreds of genes demonstrated differences in transcript usage. In a contrasting situation, the overexpression of adenosine A3 receptor (ADORA3) modifies the expression of over 1,000 genes, the number of gDTUs is still fewer than 1,000. Furthermore, DEGs associated with the overexpression of ADORA3 are significantly involved in RNA splicing, while the gDTUs exhibit a notable association with various cancers and their related biological pathways. Our findings offer valuable insights into alterations in gene expression and alternative splicing that can establish a solid groundwork for analyzing gene expression data from stable cell lines derived from HEK293T.
