Mice were given 48 tobaccos per day (4 hour), 6 days per week for 2 months or 6 months, respectively, and then samples were collected for subsequent experiments. At the same time, an air control group was maintained in a clean air environment. Whole transcriptome analysis (WTA) and targeted transcriptome analysis (TTA) combined with TCR-seq of FCAS enriched live T cells (airway, air-par, parenchyma, spleen) were performed using the BD Rhapsody Single-Cell Analysis System (BD, Biosciences). Cells were pooled from four mice per sample, with 3 parallel replicates (n = 3). |
