This project aims to enhance the anti-tumor efficacy of adoptive T-cell therapies by combining mild heat treatment (39°C) with Traditional Chinese Medicine (TCM) injections and monomeric compounds. Utilizing Jurkat T-cells as a model, the research systematically screened over 10 clinical-grade TCM injections and more than 80 monomeric compounds, identifying three injectable solutions (Shengmai Injection, Shenqi Fuzheng Injection, and Kanglaite Injection) and over 20 effective monomers (Ginsenoside Rg3, Rb1, Re, and Schizandrin A) that significantly enhanced T-cell proliferation and tumor cytotoxicity under heat-treatment conditions. High-throughput transcriptomics (HiMAP), untargeted metabolomics (UHPLC-MS/MS), and DIA-based proteomics analyses were integrated to clarify the underlying mechanisms, highlighting key pathways such as Eicosanoid signaling, PIP3-AKT activation, and regulation of apoptosis. This innovative approach combining hyperthermia and traditional Chinese medicine provides theoretical insights and a practical foundation for enhancing T-cell effectiveness, potentially improving outcomes in cancer immunotherapy. |
