To explore the influence of RAX2 on human retinal development, the CRISPR/Cas9 system was employed to disrupt critical exons of RAX2 in human embryonic stem cells (hESCs) line H9. WT and RAX2 knockout (RAX2-KO) hESCs were grown in 3D culture for 66 days (D66) using BMP4-induced Rentila Organoids (ROs) self-organization protocol, and twenty-four ROs were harvested for scRNA-seq from each of WT and RAX2-KO group. |
