Antimicrobial-resistant bacteria pose an urgent threat to human health, and the development of new antimicrobial agents is not keeping pace with the emergence of these bacteria. Therefore, exploring combination therapies of existing antimicrobial agents is one of the best solutions to tackle drug-resistant bacterial infections. To obtain a substitute for trimethoprim in the long-term used combination co-trimoxazole, we found that vitamin C could synergize with sulfamethoxazole at the minimum inhibitory concentration (MIC) level and reverse co-trimoxazole resistance in clinical isolates. Notably, a synergistic bactericidal effect was also observed between vitamin C and sulfamethoxazole in vitro and in vivo, which was independent of the synergistic effect at the MIC level. Similar synergistic effects also existed between vitamin C and trimethoprim or existing antimicrobial drugs targeting bacterial protein synthesis. |
