Pathogenic LINE-1 (pL1HS) retrotransposons promote LUSC progression by fostering immune evasion, but the mechanistic link between retrotransposon activity and TIME suppression remains unclear. We found that pL1HS drives immune suppression via liquid-liquid phase separation (LLPS)-mediated DNA sensing. Integrating multi-omics analyses-including bulk and single-cell RNA-seq with spatial proteomics-we reveal that pL1HS+ LUSC exhibits hyperactivated cGAS-STING signaling and a myeloid-derived suppressor cells (MDSCs)-rich TIME. |
