At the age of two weeks, subretinal injections were performed on the mice under a stereomicroscope. Each eye of a mouse was injected with 1 μL of scAAV8.sup-tRNA at the dose of 1x109 GC/eye. 12 weeks post injection, the eyes were enucleated and retina/RPE/choroidal/sclera complex were dissociated and immediately frozen in liquid nitrogen and ground to power in liquid nitrogen with a mortar and pestle, then dissolved in 400μL of lysis buffer. |
