Retinal degenerative diseases such as retinitis pigmentosa lack effective therapies capable of restoring vision. Photoreceptor precursor cell (PPC) transplantation has been identified as a potential solution to this issue. This study utilized PPCs efficiently derived from human induced pluripotent stem cells (hiPSCs). The use of AM580, a chemically stable retinoid analog, promoted the differentiation of hiPSCs into CRX-positive and LHX4-positive PPCs with 99.9 percent purity, ensuring the absence of off-target lineages. The retinal protective effects of these PPCs were demonstrated in an N-Methyl-N-nitrosourea-induced retinitis pigmentosa model, showing enhanced retinal intensity and spatial preservation. Donor PPCs expressed rod- and cone-specific markers and developed CtBP2-positive presynaptic specializations in close proximity to host bipolar cells. These results support the therapeutic potential of hiPSC-derived PPCs for retinal degeneration. |
