| 描述信息 | Recent clinical application with chimeric antigen receptor-modified T (CAR T) cells showed great success in treating B-cell hematological malignancies, whereas limit efficacy in solid tumors due to several obstacles such as antigenic heterogeneity. The ability of DNA demethylation reprogramming CAR T (dCAR T) cells to promote to anti-tumor activity against hematological malignancies is recognized in our previous study, but strategies to understand their potential in solid tumors remain to be confirmed. Here, we described a mechanism that dCAR T cells had the ability to eliminate heterogenous solid tumors. dCAR T cells exhibited enhanced anti-tumor ability against solid tumors in vitro and in immunocompetent syngeneic models. We showed that dCAR T cells remodeled the tumor microenvironment and importantly promoted antigen spreading of endogenous T cells to eradicate heterogenous solid tumors. It is of significant importance to note that the emergence of immunological cell death on tumor cells elicited by IFN-r expressed by dCAR T cells may recruit and activate dendric cells, as a prerequisite to promote antigen spreading. | ||||||||||||||||||||||||
